Die Rolle von Nitrilase 4 im Cyanidstoffwechsel von (L.) Heynh.
Die Detoxifikation des Atmungsgiftes Cyanid, das in Höheren Pflanzen als Nebenprodukt der Ethylenbiosynthese entsteht, beginnt mit der Bildung der Aminosäure -Cyanoalanin (-CA) aus Cystein und Cyanid. Der Abbau von -CA wird durch Nitrilase 4 (NIT4) katalysiert, die es zu Asparagin bzw. Aspartat umsetzt. Zunächst wurden durch die Untersuchung von zwei Asparaginasen Redundanzen bei dieser Reaktion in Arabidopsis ausgeschlossen. Die Analyse der NIT4-Expression erfolgte mittels -Glucuronidase-Reportergenpflanzen, die Bestimmung der intrazellulären Lokalisation von NIT4 durch transiente Expression eines NIT4-GFP-Fusionsproteins. Zur Untersuchung der Umsetzung von -CA wurden transgene Pflanzenlinien hergestellt, die eine veränderte NIT4-Expression aufwiesen (knockout- und Überexpressionspflanzen). Diese wurden physiologisch und molekular analysiert, wodurch die Beteiligung von NIT4 am Abbau von -CA in Arabidopsis gezeigt werden konnte.The detoxification of toxic cyanide, which is formed as a byproduct of ethylene biosynthesis in higher plants, starts with the formation of the amino acid -cyanoalanine (-CA) from cysteine and cyanide. The degradation of -CA to asparagine and aspartate, respectively, is known to be catalysed by Nitrilase 4 (NIT4) . To exclude redundancies in this reaction, the two asparaginases encoded in the Arabidopsis genome were overexpressed in and tested for their activity with -CA. The analysis of NIT4-expression was performed with -glucuronidase reportergene plants. Using NIT4 GFP fusion proteins in a transient expression assay the intracellular localisation of NIT4 was revealed as cytosolic. After the creation of transgenic NIT4-knockout and -overexpression lines the conversion of -CA in these plants was analysed on a physiological and molecular basis. The resulting data proved the involvement of NIT4 in the in vivo degradation of -CA
